Growth inhibition and induction of apoptosis in endometrial cancer cell line by Spatholobus suberectus Dunn

Abstract
Purpose :This study was aimed to investigate the anti-cancer effect of the stem of Spatholobus Subrectus Dunn on the human uterine endometrial cancer cells and the mechanism of Spatholobus Subrectus Dunn on endometrial cancer cell line

Methods :We counted the number of death cells treated with indicated concentration of the stem of Spatholobus Subrectus Dunn. Furthermore, flow cytometry analysis and DNA fragmentation assay were used to see whether Spatholobus Subrectus Dunn induced anti-proliferation effect was due to apoptosis and then we observed the differential cell cycle-related gene expression by western blot analysis.

Results :1. The inhibitory effect on the growth of human endometrial cancer cell treated with the stem of Spatholobus Subrectus Dunn was increased in a concentration proportional. 2. The result of flow cytometry analysis, subG1 phase arrest was investigated 35.41% in endometrial cancer cells treated with the stem of Spatholobus Subrectus Dunn and showed the fashion of proportional concentration. 3. The gene expression of p21 and Bax protein was increased according to increasing concentration but p27, p53, CDk2, cyclin E and Bcl-2 were not exchanged. 4. The character of apoptosis, DNA fragmentation was significantly observed the fashion of proportional concentration. 5. The expression of pro-caspase3 and PARP were decreased dependent on treatment concentration.

Conclusion :This study showed that the stem of Spatholobus Subrectus Dunn have the inhibitory effect on the proliferation of human uterine endometrial cancer cell and the effect was related with apoptosis.
